Pipetting Table problems

 

1. Set up a pipetting table for 3 PCR tubes so that each contains 15ul master mix, primers at 0.16uM (from 100uM), and 2, 4, or 8ng of DNA (from 280ug/ml).   Final PCR volume will be 50ul.

 

2. Set up a pipetting table for 4 PCR reactions at 40ul each so that each contains 10ul master mix, primers at 0.2uM (from 10uM), 1ng of DNA (from 65ug/ml), and 2, 4, 6, or 8mM MgCl2 (from 25mM).

 

3. Set up a PCR table to test whether Potassium has an effect on PCR.  Test 0, 1, 5, 10mM KCl (from a 1M KCl solution) along with 25ul master mix, 5ng of DNA (150ng/ul), and 0.12uM each primer (10uM) in a total PCR volume of 50ul.
